This antibody recognizes a protein of 21 kDa, identified as tissue inhibitor of metalloproteinases-2 (TIMP-2). It is closely related to TIMP-1 and shows the highest binding affinity to both the latent (pro) and active forms of 72 kDa Type IV collagenase (also known as MMP-2 or gelatinase A). It also has affinity for the active form of 92 kDa Type IV collagenase (also known as MMP-9 or gelatinase B). Interleukin 10 (IL10) is a cytokine produced primarily by monocytes and to a lesser extent by lymphocytes. This cytokine has pleiotropic effects in immuno-regulation and inflammation. It down-regulates the expression of Th1 cytokines, MHC class II antigens, and costimulatory molecules on macrophages. It also enhances B cell survival, proliferation, and antibody production. This cytokine can block NF-kappa B activity, and is involved in the regulation of the JAK-STAT signaling pathway. Knockout studies in mice suggested the function of this cytokine as an essential immuno-regulator in the intestinal tract. This antibody recognizes a protein of 75 kDa, identified as mu heavy chain of human immunoglobulins. It does not cross-react with alpha (IgA), gamma (IgG), epsilon (IgE), or delta (IgD), heavy chains, T-cells, monocytes, granulocytes, or erythrocytes. Monomeric IgM is expressed as a membrane bound antibody on the surface of B cells and as a pentamer when secreted by plasma cells. IgM antibody is prominent in early immune responses to most antigens. Aberrant levels are associated with immune deficiency states, hereditary deficiencies, myeloma, Waldenstrom's macroglobulinemia, chronic infection and hepatocellular disease. This antibody recognizes a protein of 72 kDa, which is identified as MMP2. The matrix metalloproteinases (MMP) are a family of peptidase enzymes responsible for the degradation of extracellular matrix components, including collagen, gelatin, Fibronectin, Laminin and proteoglycan. Galectin-1 is a member of the beta-galactoside-binding family and is a dimeric protein of 14 kD participating in a variety of normal and pathological processes, including cancer progression. Galectin-1 can affect the proliferation of normal and malignant cells. Inhibition of cell growth is observed in a lactose-dependent manner as lower concentrations of the lectin stimulate cell proliferation. Galectin-1 may also be implicated in the induction of apoptosis of activated T cells through the binding of exogenous galectin-1 to CD45 molecules present on the surface of lymphocytes. Recognizes the D1 domain of an N-glycosylated glycoprotein of 120 kDa with intra-chain disulfide bonds, identified as CD50 or ICAM-3 (Leucocyte Workshop VI). CD50 is the major ligand for LFA-1 (CD11a/CD18) and may have signaling role to increase adhesion. It is expressed on thymocytes and T lymphocytes and is resistant to treatment with phosphatidylinositol (PI) phospholipase C. This MAb is excellent for staining of formalin/paraffin tissues. Chromogranin A is present in neuroendocrine cells throughout the body, including the neuroendocrine cells of the large and small intestine, adrenal medulla and pancreatic islets. It is an excellent marker for carcinoid tumors, pheochromocytomas, paragangliomas, and other neuroendocrine tumors. Co-expression of chromogranin A and neuron specific enolase (NSE) is common in neuroendocrine neoplasms. Reportedly, co-expression of certain keratins and chromogranin indicates neuroendocrine lineage. The presence of strong anti-chromogranin staining and absence of anti-keratin staining should raise the possibility of paraganglioma. The co-expression of chromogranin and NSE is typical of neuroendocrine neoplasms.
